Have your headset turned on before turning MGS4 on, so it has your mic picked up. Then when you get on MGO, go to Personal Data >> Gameplay Options >> Sound, and turn Voice Chat Audio Output Device to USB/Bluetooth. Then when you are in game, hold select until you see the Voice Chat box pop up to the left of the screen. It will then be open mic, and you can turn it back off the same way by holding select.
